背景介紹：Transcription regulatory factor that mediates signaling by type I IFNs (IFN-alpha and IFN-beta). Following type I IFN binding to cell surface receptors, Jak kinases (TYK2 and JAK1) are activated, leading to tyrosine phosphorylation of STAT1 and STAT2. The phosphorylated STATs dimerize, associate with IRF9/ISGF3G to form a complex termed ISGF3 transcription factor, that enters the nucleus. ISGF3 binds to the IFN stimulated response element (ISRE) to activate the transcription of interferon stimulated genes, which drive the cell in an antiviral state.

石蠟切片免疫組化染色步驟

1、三步法（以SP試劑盒為例）

- 石蠟切片脫蠟至水

- 蒸餾水沖洗，PBS浸泡5分鐘，如果采用抗原修復，可在此步后進行

- 3% H2O2 室溫孵育5~10分鐘，以消除內源性過氧化物酶的活性，PBS沖洗，2分鐘×3次

- 5~10%正常山羊血清封閉，室溫孵育10分鐘。傾去血清，勿洗，滴加適當比例稀釋的一抗或一抗工作液，37℃孵育1~2小時或4℃過夜

- PBS沖洗，2分鐘×3次

- 滴加適當比例稀釋的*標記二抗（1%BSA-PBS稀釋），37℃孵育10~30分鐘；或滴加第二代*標記二抗工作液，37℃或室溫孵育10~20分鐘

- PBS沖洗，2分鐘×3次

- 滴加適當比例稀釋的辣根酶標記鏈霉卵白素（PBS稀釋），37℃孵育10~30分鐘；或第二代辣根酶標記鏈霉卵白素工作液，37℃或室溫孵育10~20分鐘

- PBS沖洗，2分鐘×3次

- 顯色劑顯色（DAB或AEC）

- 自來水充分沖洗，復染，脫水透明、封片

- 如果必要，可選用avdin封閉內源性*
