The microtiter plate provided in ELISA Kit has been precoated with an antibody specific to the target protein. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated specific polyclonal antibody preparation,and Avidincon jugated to Horseradish Peroxidase（HRP） is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain target protein, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of the samples is then determined by comparing the O.D. of the samples to the standard curve.

3. 蒸餾水或去離子水，全新濾紙 標(biāo)本的采集及保存
1. 血清：全血標(biāo)本請于室溫放置 2 小時或 4℃過夜后于 1000 x g 離心 20 分鐘，取上清即可
檢測，或?qū)?biāo)本放于-20℃或-80℃保存，但應(yīng)避免反復(fù)凍融。
2. 血漿：可用 EDTA 或肝素作為抗凝劑，標(biāo)本采集后 30 分鐘內(nèi)于 2 - 8° C 1000 x g 離心 15
分鐘，或?qū)?biāo)本放于-20℃或-80℃保存，但應(yīng)避免反復(fù)凍融。
3. 細(xì)胞培養(yǎng)物上清或其它生物標(biāo)本：請 1000 x g 離心 20 分鐘，取上清即可檢測，或?qū)?biāo)本
放于-20℃或-80℃保存，但應(yīng)避免反復(fù)凍融。
注：以上標(biāo)本置 4℃保存應(yīng)小于 1 周，-20℃或-80℃均應(yīng)密封保存，-20℃不應(yīng)超過 1 個月，-80℃
不應(yīng)超過 2 個月；標(biāo)本溶血會影響zui后檢測結(jié)果，因此溶血標(biāo)本不宜進行此項檢測。
樣本處理：血清或血漿標(biāo)本*稀釋 10 倍，如：稀釋 10 倍，取 100uL 血清或血漿加入 900uL
樣品稀釋液。建議各實驗室在操作前*行預(yù)實驗以建立*稀釋倍數(shù)。
