周期素依賴性激酶4抗體,Anti-CDK4抗體產品介紹：The protein encoded by this gene is a member of the Ser/Thr protein kinase family. This protein is highly similar to the gene products of S. cerevisiae cdc28 and S. pombe cdc2. It is a catalytic subunit of the protein kinase complex that is important for cell cycle G1 phase progression. The activity of this kinase is restricted to the G1-S phase, which is controlled by the regulatory subunits D-type cyclins and CDK inhibitor p16(INK4a). This kinase was shown to be responsible for the phosphorylation of retinoblastoma gene product (Rb). Mutations in this gene as well as in its related proteins including D-type cyclins, p16(INK4a) and Rb were all found to be associated with tumorigenesis of a variety of cancers. Multiple polyadenylation sites of this gene have been reported.

抗原修復的幾種常用方法（供參考）
1、 微波爐加熱：
在微波爐里加熱0.01*緩沖液（pH6.0）至沸騰后將組織芯片放入，斷電，間隔5-10分鐘，反復1-2次。根據玻片情況可參考采用微波加熱“3-5-3法”3分鐘溫火沸騰后靜止5分鐘，再溫火沸騰3分鐘即可。
適用的抗原：來源于人、大鼠、小鼠的組織標本；來源于其他動物種屬的組織標本。
2、 沸熱修復：
電爐或水浴鍋加熱0.01M*緩沖液（pH6.0）至95℃左右，放入組織芯片加熱10-15分鐘。
3、 高壓熱修復：
在沸水中加入EDTA（pH8.0）或0.01m*緩沖溶液（pH6.0）.蓋上不銹鋼鍋蓋，但不能鎖定。將玻片置于金屬染色架上，緩慢加壓，使玻片在緩沖液中浸泡五分鐘，然后將蓋子鎖定，小閥門將會升起來。10分鐘后除去熱源，置入涼水中，當小閥門沉下去后打開蓋子。此方法適用于較難檢測或核抗原的抗原修復。（骨及軟骨組織的標本選擇較溫和的微波加熱修復） 。
4、 酶消化方法：
常用0.1%*和0.4%*液。*使用前預熱37℃，消化時間約為5-30分鐘。
適用于被固定遮蔽的抗原：包括 Collagen.GFAP. Complement.Cytokeratin.c-erB-2.LCA.LN.等。

修復液：0.01M 枸櫞酸（pH6.0）或者0.05M EDTA（pH8.0）
抗體修復方法：
方法1：沸水浴修復，將盛有修復液和玻片的燒杯置于沸水浴環境，保持外部沸騰狀態15min，自然冷卻至室溫；
方法2：微波修復，將盛有修復液和玻片的燒杯置于微波爐中，高火5min，停火3min，中火5min，自然冷卻至室溫。
